NEBuffer Chart

To help select the best conditions for double digests, this chart shows the optimal (supplied) NEBuffer and approximate activity in the four standard NEBuffers and NEBuffer EcoRI for each enzyme. In addition, the table also shows recommended reaction temperature, heat inactivation temperature, recommended diluent buffer and whether the enzyme is Time-Saver qualified (i.e., cleaves substrate in 5 minutes).

Double Digest Finder

Cleaving a DNA substrate with two restriction enzymes simultaneously (double digestion) is a common timesaving procedure. Use this tool to select reaction conditions amenable to any two NEB restriction enzymes.

DNA Sequences and Maps

NEBcutter

This tool will take a DNA sequence and find the large, non-overlapping open reading frames using the E.coli genetic code and the sites for all Type II and commercially available Type III restriction enzymes that cut the sequence just once. It also shows the enzymes which could be used in a complete digest to excise each open reading frame that it finds. From the initial display there are options to go further, including custom digestion with enzymes of your choice and various displays of the digest.
